چکیده

Background Excessive proliferation and migration of vascular smooth muscle cells (VSMCs) are associated with the pathogenesis atherosclerosis (AS). Eplerenone (EPL), a novel selective aldosterone receptor blocker, plays substantial role in treatment cardiovascular disease. The G protein-coupled estrogen (GPER) is target EPL as STITCH website predicated. Objectives We aimed to investigate roles AS identify its potential mechanisms action. Material methods Oxidized low-density lipoprotein (ox-LDL) was employed stimulate VSMCs establish cellular model AS. ability cell examined using Cell Counting Kit-8, expression proliferation-related proteins tested immunofluorescence staining western blot analysis. Subsequently, migration-associated were evaluated wound healing assay, transwell assay Then, GPER determined analysis absence or presence EPL. To explore regulatory ox-LDL-stimulated VSMCs, overexpressed, followed by measurement migration. Results Ox-LDL stimulation notably upregulated expression, whereas downregulated dose-dependent manner. Additionally, markedly inhibited highest dose resulted most marked effect. By contrast, overexpression reversed inhibitory effects on VSMCs. Conclusions suppressed ox-LDL-induced partly through downregulation GPER, providing new mechanism support for use clinical

The Effect of Adiponectin on Osteonectin Gene Expression by Oxidized Low Density Lipoprotein-Treated Vascular Smooth Muscle Cells

Osteonectin is a bone- associated protein involved in vascular calcification. Adiponectin may protect against cardiovascular disease but possible effects on vascular calcification have been poorly studied. The aim of this study was to investigate the modulatory effect of adiponectin on oxidized low density lipoprotein (oxLDL)- induced expression of osteonectin in human aorta vascular smooth mus...

Effect of Oxidized Low Density Lipoprotein on the Expression of Runx2 and SPARC Genes in Vascular Smooth Muscle Cells

Background: Vascular calcification is an important stage in atherosclerosis. During this stage, vascular smooth muscle cells (VSMC) synthesize many osteogenic factors such as osteonectin (encoded by SPARC). Oxidative stress plays a critical role in atherosclerosis progression, and its accumulation in the vascular wall stimulates the development of atherosclerosis and vascular calcification. The...

Tanshinone IIA inhibits AGEs-induced proliferation and migration of cultured vascular smooth muscle cells by suppressing ERK1/2 MAPK signaling

Objective(s): Vascular smooth muscle cells (VSMCs) play a key role in the pathogenesis of diabetic vascular disease. Our current study sought to explore the effects of tanshinone IIA on the proliferation and migration of VSMCs induced by advanced glycation end products (AGEs). Materials and Methods: In this study, we examined the effects of tanshinone IIA by cell proliferation assay and cell mi...
